چکیده مقاله:

Astroblastoma(AB) is a poorly defined central nervous system (CNS) tumor, included along with polar spongioblastoma and gliomatosis cerebri in the group of neuroepithelial tumors of uncertain origin in the June ۲۰۱۶ World Health Organization (WHO) Classification of tumors of the CNS. They are rare neoplasms that affect primarily patients of young ages.[۱] The exceptional growth of information and advances in technology in biomedical fields has been overwhelming for labratorary scientists over the last decade. Therefore, research has been moving from being strictly conducted in a real life lab environment to a ‘virtual lab’ environment where management and analysis of data are done[۲]. Initially, gene expression data of AB patients (GSE۱۶۵۸۱۳) was obtained from the NCBI Gene Expression Omnibus (GEO) and then analyzed by GEO۲R to find differentially expressed genes (DEGs).Through UniProt[۳] and GeneCards[۴], gene ontology information and biological pathway involvement were understood. Furthermore, miRWalk[۵], were utilized to find significant miRNA-mRNA interactions in the ۳’UTR region. Additionally, the selected miRNA was searched in lncbasev۳ [۶] to find strong interactions with lncRNAs and construct a predictive LncRNA-miRNA network. Through analysis of the GEO dataset, a gene named EGR۱was found to be considerably upregulated (|logFC| = ۲.۳۹۱۳ , adj. P value =۱.۰۹e-۱۰) in AB samples.The transcription factor EGR۱ (early growth response۱) is a member of zink finger transcription factor family which plays an essential role in cell growth and proliferation. Analysis of possible miRNA-mRNA interactions revealed hsa-miR-۳۹۴۰-۳p as a significant interactor to EGR۱ mRNA. This miRNA was then searched in Lncbasev۳ and HSALNG۰۰۳۶۳۶۳,lnc-ELOVL۶-۳,HSALNG۰۰۳۶۳۶۵ had the strongest interactions. In conclusion EGR۱ is overexpressed in GBM and forms a possible ceRNA network among hsa-miR-۳۹۴۰-۳p , HSALNG۰۰۳۶۳۶۳,lnc-ELOVL۶-۳ and HSALNG۰۰۳۶۳۶۵ .
