The immunomodulatory effects of Curcumin in experimental autoimmune encephalomyelitis through activation of Th۲/ Treg cells immune responses

Background and aim: Multiple Sclerosis (MS) characterized by demyelination of the central nervous system, which is impacted by hereditary and environmental variables. This research will look at the cellular and molecular processes through which curcumin can influence the progression of EAE disease.Materials and method: A total of ۲۴ female C۵۷BL/۶ mice were utilized in this study. Subcutaneous injection of Myelin Oligodendrocyte Glycoprotein (MOG) and full Freund's adjuvant was used to induce EAE. Pertussis toxin was administered and on the ۲۵th day, animals were slaughtered. TGF-β, IL-۱۰, IL-۴, TNF-α, and IFN-γ were measured in spleen cells using an ELISA kit. Real-time PCR was used to analyze the mRNA levels of cytokines and transcription factors such as IL-۱۳, IL-۳۳, IL-۲۷, IL-۳۵, GATA۳, STAT۶, FOXP۳, and CTLA۴ in brains and spinal cords. The proliferation of splenocytes was measured using the BrdU test.Results: The treated group had less lymphocytic infiltration and demyelination. The treatment group's mean weight increased significantly but the disease's progression was shown to be slower in the treated group. Inflammatory cytokine release by inflammatory T-cells was dramatically reduced in the treated groups. However, the production of anti-inflammatory cytokines has increased dramatically in these groups. The proliferation of MOG-sensitive T-cells was dramatically reduced in the treated groups. The expression of transcription factors and cytokines associated with Treg and Th۲ cells rose considerably in the treated group.Conclusion: Curcumin appears to have a preventive impact on disease progression and ameliorate illness symptoms in EAE through favoring Treg and Th۲ cell proliferation and function.
