Background and aim: Neutrophil Extracellular Traps (NETs) are net-like structures composed of DNA-histone complexes and produce via a unique form of cell death called NETosis. It has already been proven that the NETs have an increased amount in the blood and tissues in a variety of cancers. In this systematic review study, we review NETs-related articles in cancers and its role in tumor genesis, tumor progression, and prognosisMaterials and method: Eligible studies by electronic search through various databases including Medline and Cochrane (from ۲۰۰۰ to ۲۰۲۱) related to neutrophil extracellular traps in cancers were examined using the following Key words: neutrophil extracellular traps, cancer and tumor.Results: Findings Of ۵۹۵ screened and of the ۲۳ studies included, ۱۴ were observational, ۲ were in vivo experimental, ۴ were in vitro, and ۳ used a combination of these approaches. NET formation in the tumor microenvironment promotes cancer progression and metastasis. Analyzes showed that NET-related proteases, including neutrophil elastase and matrix metalloproteinase ۹, by acting on cancer cells or altering their microenvironment, they awaken dormant cancer cells through ECM regeneration. NETs can also trap cancer cells and act as a substrate for cancer cells to increase metastasis. An increase in NETs is often associated with a poor prognosis and a decrease in survival.Conclusion: NET-targeted therapeutic processes have been identified as one of the research priorities in the field of cancer. However, it seems that it is still a little early to hope for such treatments. Key Words Neutrophil extracellular traps, Cancer, NETs, Tumor, Metastasis, prognosis
