Objective: The skin is known as the larges t organ of the bodyand also a barrier that protects the human agains t environmentalchanges. Burns is one of the mos t common injuries that occurto the skin. In this s tudy, we used a commercial collagen sheetwound dressing material as a subs trate for seeding of humanforeskin fibroblas ts (hFF) to create a biological wound dressing.Materials and Methods: The hFF cells were provided by theRoyan S tem Cell Bank and cultured in an appropriate medium.The bovine collagen sheet (Suprasorb® C) was cut out in dimensionsof ۰.۲×.۰۲ cm۲ and inserted into the wells of a ۲۴-wellplate. The cells were loaded at the density of ۱۰۵ cells/well onthe collagen sheet. The MTS assay was performed to analyzethe viability and proliferation of the cells on the collagen sheetafter one, two, and three days of incubation.Results: The MTS assay showed significantly higher cell viability and metabolic activity in cell-laden collagen cons tructsin comparison to the control group (cell only) after one day inculture. There were no significant differences between groupsafter two- and three- days of incubation.Conclusion: Based on the results of this s tudy, it can be concludedthat the collagen sheet with hFF cells could be consideredas a biological wound dressing. However, in vivo s tudy onan animal model of burn can provide more information aboutthe effectiveness of this biological bandage
