Objective: Cancer three-dimensional (۳D) modeling, with tissueengineering assis tance, will mimic an appropriate bodysys tem by modifying the tumor microenvironment. Exosometherapy has recently been used in cancer s tudies due to its anticancereffects. In this s tudy, the effects of crab exosomes in abreas t cancer (BC) ۲D and ۳D model were inves tigated in vivo.Materials and Methods: In this s tudy, female mice aged ۶ to ۸weeks were used. By the Chitosan/β-glycerol phosphate hydrogel(Ch/β-GP) and ۱×۱۰۶ ۴T۱ cells, a cancer ۲D and ۳D modelwere created in the mice. In both models, ۱ mg/ml crab exosomeand ۵ mg/kg doxorubicin (DOX) were injected by intratumoral(IT), intravenous (IV), and intraperitoneal (IP) methodsinto mice’s right flank in ۳ intervals. After ۲۱ days, the micewere sacrificed and the tissues were removed. Tumor weightand size were measured. VEGF, Bcl۲, and P۵۳ gene expressionlevels were assessed by real-time polymerase chain reaction(PCR). Nitric oxide (NO) secretion from the cancer ۳D modelwas evaluated by Griess assay.Results: Tumor size and weight in treated groups with exosomeand DOX were reduced significantly (P value ≤۰.۰۰۱, Pvalue ≤۰.۰۰۲, P value ≤۰.۰۲). Changes in VEGF, Bcl۲, and P۵۳gene expression levels were less in the ۳D model than in the۲D model. Decreased NO secretion was observed in all groupscompared to the control group.Conclusion: BC ۳D model was shown greater drug resis tance;this can be one of the features of a malignant tumor. Therefore,it can be used in cancer s tudies with greater confidence. Crabexosome was effective in BC inhibition
