چکیده مقاله:
Objective: Breas t cancer s tem cells are resis tance to conventionaltherapy. NK cell therapy has not been satisfactory dueto immunosuppressive of tumor microenvironment in breas tcancer. hTERT as tumor-associated antigens (TAAs) is overexpressedin ۸۵% of tumors and has a role in self-renewal, proliferationof the CSC, immortality, and tumorigenesis. Thus, inthis s tudy, we inhibited telomerase and evaluated the NK cellcytotoxicity and apoptosis of Breas t cancer s tem cells.Materials and Methods: MDA-MB-۲۳۱cells were treatedwith IC۵۰ level of BIBR۱۵۳۲. After ۲۴ hours, NK cells wereadded to the treated cells and incubated for ۵ hours. Then, NKcell cytotoxicity, Annexin V/۷AAD experiments, and expressionof apoptosis related genes were analyzed.Results: Our results showed that telomerase inhibition increases apoptosis in breas t cancer s tem cells.Conclusion: BIBR۱۵۳۲ inhibit telomerase and sensitize breas tcancer s tem cells to NK cell therapy. Therefore, Telomerase inhibitioncan be beneficial in the proper function of NK cells.
