Objective: S tem cell-based regenerative medicine tries to findnovel approaches to treating severe medical disorders includingwound. The insulin-like growth factor (IGF) family contributesefficiently to growth, cell proliferation and differentiation. AlsohUC-MSCs (human umbilical cord mesenchymal s tem cells)play important role in regenerative medicine and they can releaseinsulin-like growth factors.Materials and Methods: In this examination, ۲۰ male Wis tarrats (۲۵۰ ± ۲۰g) were randomly divided into two groups afterwound creation on the dorsal side of the animals. Group I was thecontrol group and normal saline were injected into the peripheralarea of the wound intradermal. The second group (group II)was the human umbilical cord hUC-MSCs CM (human umbilicalcord mesenchymal s tem cells conditioned medium) treatmentgroup which ۱cc Wharton´s jelly-derived hUC-MSCs CMwas injected in the peripheral area of the wound intradermally.The wound healing rate was measured and marked on the ۰th,۵th, ۱۰th and ۱۴th days of the process. Subsequently, all animalswere euthanized and the wound samples were s tored for complementaryassays. The expression level of IGF-۱ and IGF-۲were assayed by real-time polymerase chain reaction (PCR).Results: The results from the measurement of the woundssurfaces showed that the treatment was significantly effectivefor wound healing in comparison to the control group (P value<۰.۰۵). Also the expression level of IGF-۱ and IGF-۲ was significantlyincreased in compare with the other group (P value<۰.۰۵ and P value <۰.۰۰۵).Conclusion: HUC-MSCs conditioned medium therapy increasedthe expression of IGF-۱ and IGF-۲ in the wound samplesand it improved wound healing.
