Objective: Subs tituting differentiated s tem cells for los t anddamaged cells is one of the proposed therapeutic options forspinal cord injury (SCI). Human endometrial s tem cells (hEn-SCs) are a potential source of mesenchymal s tem cells that candifferentiate into various neural cell types. Many researchersare developing protocols for motor neuron differentiation to regeneratecells that match the los t endogenous ones as a possiblecure for SCI and neurodegenerative illnesses. Valproate sodium(VAS) is a small molecule with anti-inflammatory and neuroprotectivefeatures and has the ability to pass the blood-brainbarrier. Our goal was to s tudy the effect of VAS on improvingmotor neuron differentiation of human endometrial s tem cells.Materials and Methods: hEnSCs were extracted via collagenasetype I and were authenticated at passage three by flowcytometry. The motor neuron differentiation protocol containedthree phases: ۱. pre-induction phase (۲۴ hours), ۲. inductionphase (firs t week), and ۳. survival phase (second week). TheVAS (۱۰۰ μg/mL) was added from phase ۲ or phase ۳ of differentiation.Immunofluorescence (IF) was used to assess theexpression of neurofilament heavy polypeptide (NF-H) andsynaptophysin (SYP) after ۱۵ days.Results: The IF results revealed NF-H and SYP high protein expression (two important neural-specific markers) in the differentiationgroup, which VAS was added in the third phase(second week) of differentiation. In contras t, the other differentiationgroup (VAS was added in phase ۲) did not show NF-Hand SYP expression.Conclusion: This s tudy points out the importance of time-dependentadding of VAS to improve motor neuron-like cell differentiationof hEnSCs.
