Objective: Shilajit, as a herbomineral natural subs tance, hasbeen mos t widely used for the remedy of a numerous of illnesssuch as bone defects in traditional folk medicine sincehundreds of years ago. The aim of the present s tudy was toexplore the effect of Shilajit on the os teogenic differentiationof human(adipose derived mesenchymal s tem cells (ASCs) intwo- and three-dimensional cultures.Materials and Methods: ASCs were isolated and suspended in۱% (w/v) alginate solution with or without Shilajit and seededinto culture plates at a density of ۱۵ × ۱۰۴ cell per ۵۰۰μL /wellfor three-dimensional (۳D) cultures. For two-dimensional (۲D)cultures, a suspension of ASCs was seeded into culture plates ata density of ۱۵ × ۱۰۳ cell/well and treated with ۵۰۰ μg/ml Shilajit.Then, characterization was done using SEM/EDX, ALP (alkalinephosphatase) activity, Alizarin red s taining, and Ramanconfocal microscopy.Results: According to findings, Shilajit/alginate combinationin the presence or absence of os teogenic medium significantlyincreased ALP activity and mineralization, compared to the ۲Dmatched groups at all time points (P value <۰.۰۵). In addition,calcium deposition level was obviously high in Shilajit/ alginatecombination with or without os teogenic medium comparedto ۲D matched groups on days ۱۴ (P value <۰.۰۰۰۱), and ۲۱(P value <۰.۰۰۱and P value <۰.۰۱, respectively). Shilajit couldalso promote os teogenic differentiation of s tem cells with orwithout os teogenic medium in two-dimensional cultures. ButShilajit/alginate combination s tarts os teogenic differentiation ina short period of time.Conclusion: As Shilajit accelerates the differentiation of ASCsinto the os teoblas ts, without changing the physical properties ofthe alginate hydrogel, this combination may pave the way formore promising remedies considering bone defects
