The Effect of Copper Nanoparticles on In Vitro Maturationof NMRI Mouse Oocyte

Background: In vitro maturation (IVM) of oocyte is an assis tedreproductive technique that avoids the side effects of gonadotropinsand ovarian hypers timulation syndrome (OHSS).Improving the culture medium is consdidered pivotal s tageto promote oocyte maturation. Considering the importance ofcopper in mitochondrial oxidative phosphorylation and protectionagains t oxidative s tress and ATP production, the aim ofthis s tudy was to inves tigate the effect of copper nanoparticles(CuNPs) as an antioxidant in IVM medium and evaluate themitochondrial activity of immature mouse oocytes treated withexamined nanoparticles.Materials and Methods: The immature oocytes were collectedfrom ۶-۸ weeks old NMRI mice and divided to four groupswith different concentrations of nanoparticles (۰.۰۰۲%w/v,۰.۰۰۴%w/v, ۰.۰۰۶%w/v, and ۰ %w/v (as a control group) toobtain the optimal dosage. IVM rate, mitochondrial activity(JC۱ s taining assay) and oocyte survival (Trypan blue s tainingassay) were evaluated in response to the addition of CuNPs duringIVM process.Results: The nuclear maturity rate of oocytes didn’t show anysignificant difference between the control group and the differentconcentrations of CuNPs. Evaluation of oocyte survival datadid not show any significant reduction in survival rate betweenthe control group and different concentrations of CuNPs. Themitochondrial activity rate was increased by elevated concentrationsof CuNPs, although there was no significant differencebetween the control group and the maximum concentrations ofCuNPs.Conclusion: The use of CuNPs in IVM medium had no adverseeffect on the survival and maturation rate of oocytes
