Growth Factor Improves In Vitro Maturation of Domestic Cat Oocytes

Background: Domes tic cat is the only non-endangered speciesamong felids and could serve as an oocyte donor to preservethe endangered feline species. Nevertheless, in vitro maturation(IVM) is suboptimal in cats, despite major progress over recentyears. In this s tudy, we have assessed the effects of growthdifferentiation factor ۹ (GDF۹) and bone morphogenetic protein۱۵ (BMP۱۵) combination and insulin-like growth factor۱ (IGF۱) and Epidermal growth factor (EGF) combination onIVM of cat oocytes.Materials and Methods: Following spay surgery, cumulusoophorus complexes (COCs) were harves ted from ovaries andrandomly allocated to four groups including control (basic medium),IE (basic medium with ۱۰ μg/ml IGF۱ and ۱۰ μg/mlEGF), GB (basic medium with ۲۰۰ ng/ml GDF۹ and ۲۰۰ ng/mlBMP۱۵) and IE+GB (basic medium with ۱۰ μg/ml IGF۱ and۱۰ μg/ml EGF plus ۲۰۰ ng/ml GDF۹ and ۲۰۰ ng/ml BMP۱۵)for IVM at ۳۸ °C for ۲۴hours. Maturation and degeneration ofoocytes were evaluated by s tereomicroscope.Results: The proportion of matured oocytes in IE (۱۳/۷۳:۱۷.۱۱%), GB (۹/۶۵: ۱۳.۸۵%), and IE+GB (۸/۵۸: ۱۳.۷۹%)groups was greater than that in the control group (۲/۷۳: ۲.۷۴%;P<۰.۰۵). IGF۱ and EGF combination positively impacted felineoocytes IVM (P=۰.۰۳); however, GDF۹ and BMP۱۵ combinationdid not influence IVM rate (P>۰.۰۵). Degeneration ratedid not differ among the control (۱۶/۷۳: ۲۱.۹۲%), IE (۱۲/۷۳:۱۶.۴۴%), GB (۱۶/۶۵: ۲۴.۶۲%) and IE+GB (۱۱/۵۸: ۱۸.۹۷%)groups (P>۰.۰۵).Conclusion: In conclusion, the present s tudy showed incorporationof IGF۱ and EGF into IVM medium could enhance thematuration rate of feline oocytes.
