Background: Curcumin exerts beneficial therapeutic effects.However, chronic and high dose consumption of curcuminis able to exert deleterious side effects on male reproductivesys tem. The curcumin-related effects have been improved byconverting the curcumin to nano-micelle form named as nanomicellecurcumin, which is able to alter its beneficial or deleteriouseffects. In the current s tudy, Wis tar male rats were usedto uncover the possible dose-dependent effect of nano-micellecurcumin on spermatogenesis, intracellular metabolomics, andautophagy process.Materials and Methods: For this purpose, ۲۴ male rats (۲۰۰ ±۲۰ g) were randomly divided into control, ۷.۵ mg/kg, ۱۵ mg/kg,and ۳۰ mg/kg nano-micelle curcumin-received (orally) groups.Following ۴۸ days from nano-micelle curcumin adminis tration,the his tomorphometric, his tochemical (intracytoplasmic carbohydrateand lipid accumulation in germ and somatic cells), andthe expression levels of Atg۷, Beclin-۱, p۶۲, LC۳-I in the testiculartissue have been evaluated.Results: Observations revealed that nano-micelle curcumin, ina dose dependent manner, could decrease the Leydig and Sertolicells number, germinal epithelium’s height, tubular diameter,and the percentages of seminiferous tubules with positive tubulardifferentiation (TDI) and spermiogenesis (SPI) comparedto the control group. Furthermore, the a apoptotic Leydig cellsnumber was increased (P<۰.۰۵) and the number of Leydig cellswith intracytoplasmic carbohydrate accumulation and s teroidogenicactivity were dose-dependently decreased in the nano-micelle curcumin-received groups (P<۰.۰۵).In comparisonwith control rats, the total number of Sertoli cells containingintracytoplasmic carbohydrate was decreased and the numberof apoptotic Sertoli cells as well as cells with high accumulationof lipid droplets were increased in the nano-micelle curcuminreceivedgroups (P<۰.۰۵). Finally, the nano-micelle curcuminreceivedgroups exhibited a remarkable (P<۰.۰۵) increment inthe expression levels of autophagy-related genes.Conclusion: Our data shows that overdose consumption of thenano-micelle curcumin (ignoring its appropriate pharmacologicaldose) is able to up-regulate the autophagy-related genes expressionby altering the metabolic interactions in the tes ticulartissue.
