Background: Nandrolone decanoate (ND) adminis trationhas been reported to significantly induce germ cell apoptosis,suppress tes ticular endocrine s tatus, and decrease the spermmorphology and motility ratio. On the other hand, moderateintensityexercise training protocol (MICT) is shown to boos tanti-apoptotic proteins expression, sys temic and tes ticularendocrine s tatuses, and sperm quality. The current s tudy wasaimed to inves tigate the protective effect of MICT on ND-induceddetrimental effects on sperm parameters by exploringpossible changes in β-defensin (۱-۴) expression and tes ticularantioxidant s tatus.Materials and Methods: For this purpose, ۲۴ mature maleWis tar rats were divided into sedentary control, MICT (E-sole),ND-adminis trated (۱۰ mg/kg, ۳ times/week, for ۴۰ days, ip),and E+ND-induced groups. Following ۴۰ days, the tes ticularoxLDL and carbonyl groups (CG, markers for lipid and proteinperoxidation), β-defensins (۱-۴) expression levels, and spermcount, motility, and DNA integrity were examined and comparedbetween groups.Results: Observations revealed that ND could significantly(P<۰.۰۵) increase tes ticular oxLDL and CG versus control rats.Moreover, it could remarkably (P<۰.۰۵) suppress the epidydimalβ-defensins (۱-۴) expression and decrease the spermcount, motility, and DNA integrity. However, >>exercise trainingcould diminish tes ticular oxLDL, CG, and boos t β-defensins(۱-۴) expression, and ameliorate sperm count, motility, andDNA integrity (P<۰.۰۵).Conclusion: Minding that tes ticular antioxidant s tatus cooperateswith β-defensins to protect sperm motility, nuclear maturation,and fertilization potential, we can conclude that exercisecould improve sperm motility and DNA integrity by inhibitingoxidants-induced lipid and protein peroxidation in tes ticles andβ-defensins (۱-۴) expression level in the epididymis.
