Objectives:The human semen contains a heterogeneous population of Extracellular vehicles (EVs) have been consistently implicated in promotion of sperm motility, regulation of sperm capacitation, induction of acrosome reaction, or antibacterial and antiviral mechanisms. The purposes of this study investigate the seminal plasma exosomes on sperm function sperm function in oligoasthenoteratozoospermia patients.Methods:Normozoospermic, and severe oligoasthenoteratozoospermia (men۲۵–۴۰years of age) were considered for the study. Seminal plasma was collected and processed to separate spermatozoa and exosomes. Exosome uptake by spermatozoa was monitored by means of Bradford and flow cytometry. The effect of exosomes on spermatozoa was determined by evaluating sperm parameters According to the World Health Organization criteria (WHO) (۲۰۱۰(.Results:We isolated and characterized exosomes from seminal plasma of Normozoospermic, severe oligoasthenoteratozoospermia patients. Exosomes concentration derived from oligoasthenoteratozoospermia significantly was decreased than normozoospermic individuals (p < ۰.۰۵). The current study showed a statistically significant highly positive correlation in Exosomes concentration with sperm count, motility, normal morphology, and viability. However, it shows significant negative correlation with sperm DNA fragmentation (p < ۰.۰۵). Conclusions:These findings provide evidence that exosomes in seminal plasma can effect on sperm quality. In the future, exosomes assessment could serve as a diagnostic tool to ensure the sperm parameters infertile couples with male factors.
