Frequency of cagE and cagM genes in Helicobacter pyloriisolated from patients with digestive disorders

Background and Aim : Helicobacter pylori is considered one of the most important humanpathogenic bacteria that has infected almost half of the world's population, which can cause alifelong infection if not treated. However, the amount of pollution varies greatly according to thegeographical area. The occurrence of infection and the outcome of the disease depend on variousfactors, including the pathogenic factors of bacteria, the conditions of the host and theenvironment. The aim of this study is to determine the frequency of CagE and cagM genes instrains isolated from gastric biopsy samples of a number of patients referred to medical centers.Methods : In this research, ۳۵۰ patients who suffered from gastrointestinal diseases were studied.DNA extraction of all biopsy samples was done using chloroform method and then using PCRmethod and glmM specific primer, samples containing Heliobacter pylori were identified. Also, inorder to identify cagM and cagE genes, primers extracted from previous studies and PCR techniquewere used. At the end, the duplex PCR reaction products were electrophoresed on ۱% agarose geland the results were observed by Gel Documentation device.Results : From a total of ۳۵۰ biopsy samples from patients with an average age of ۴۹ years, ۱۹۰women and ۱۶۰ men, ۱۲۳ Helicobacter pylori samples were positive based on PCR and glmMgene. The prevalence of cagE gene was ۵۶% and cagM was ۵۴% and ۴۳% for both genes.Conclusion : The frequency of this bacteria was ۲۶% using urease test and ۳۵% using PCRmethod. According to the results of previous studies, the higher specificity of molecular tests inidentifying this bacterium can be pointed out. In the present study, the frequency of cagE gene was۵۶%. cagE can be used as a marker for healthy cag-PAI samples. R. Mattar and others reportedthat the cagM gene is a marker to identify people at higher risk of peptic ulcer disease in Brazil.The simultaneous presence of the two genes studied in the research It was also observed in ۴۳%of the samples, and in general, the results of this study are consistent with the results of previousstudies.
