چکیده مقاله:

Background and Aim : Shigella SPP. is an invasive Gram-negative, facultative anaerobe bacilliof the family Enterobacteriaceae which infects millions of people annually and is characterized bywatery or bloody diarrhea, cramping, fever, and dehydration. Globally, Shigella infection causessignificant mortality and morbidity, nearly ۱۶۷ million Shigella episodes annually are estimatedand most of these cases are related to low income countries. Shigellosis is more common inchildren under five years old. One of the most invasive shigella spp. with the high distributionamong low income countries is Shigella flexneri. Investigating the invasion of Shigella speciescan be effective in better understanding of this bacterium and improving the treatment process ofthe disease caused by it. One of the methods of investigating the pathogenicity of organisms is theuse of cell culture. Therefore, we decided to evaluate the invasion of Shigella flexneri in Hela cellsusing the plaque formation assay.Methods : In this study, ۷ virulence genes including ipaH, ipaB, ipaC, ipaD, virA, ipgD and senwere investigated in five Shigella strains using PCR. Invasion of Shigella species was evaluatedusing the plaque formation assay technique in Hela cells. The diameter and number of formedplaques were analyzed using ImageJ software.Results : Strain number ۲ was recognized as the most invasive strain by having all ۷ virulencegenes and forming ۳۶۰ plaques. Strain number ۵ contains ۴ virulence genes and showed the leastinvasion by forming ۲۳ plaques. The virulence profile and the number of plaques are as follows:SF۱: ipaH,ipaB,ipaC,ipaD,virA/۱۳۴ SF۲: ipaH, ipaB, ipaC, ipaD, ipgD, sen, virA / ۳۶۰ SF۳: ipaH,ipaB, ipaC, ipaD, ipgD, sen, virA / ۱۳۸ SF۴: ipaH, ipaB, ipaC, ipaD, ipgD, sen, virA / ۶۵ SF۵:ipaH, ipaB, ipaC, virA / ۲۳Conclusion : Most species of Shigella flexneri have a high invasion power and can destroy theintestinal epithelium. The reason for the invasiveness of this bacterial species is the diversity of itsvirulence factors.
