Background and Aim : Following the COVID-۱۹ pandemic, numerous studies to clarify the mainreason for the discrepancy in susceptibility to COVID-۱۹ among populations are undergoing. Hostgenetic changes like single nucleotide polymorphisms (SNP) is one of the main factors thatinfluence the patients susceptibility to viral infectious diseases. This study aimed to investigate theassociation between host genetic polymorphisms of Toll-like receptor۷ (TLR۷) gene that Involvedin the immune system and susceptibility to COVID-۱۹ disease in a sample of the Iranianpopulation.Methods : This study is a retrospective case-control study which in ۲۴۴ and ۲۱۵ patients withCOVID-۱۹ disease (positive confirmed with molecular test) as case group and ۱۵۶ and ۱۲۲suspected patients to COVID-۱۹ disease (negative confirmed with molecular test) as control groupwere genotyped for TLR۷ polymorphism TLR۷: rs۱۷۹۰۰۸ A/T and rs۱۷۹۰۰۹ T/C respectively.Studied population selected from Ghods clinic affiliated to Tehran Medical Sciences, Islamic AzadUniversity, Tehran. Genomic DNA was extracted from nasopharyngeal and oropharyngealspecimens and genotyping of gene polymorphisms were performed using Polymerase ChainReaction Restriction Fragment Length Polymorphism (PCR-RFLP) method.Results : In the male gender AA, AT and TT genotypes distribution in TLR۷ rs۱۷۹۰۰۸ were۵۳.۶%, ۳۶.۶% and ۹.۸% in case group and ۷۶.۳%, ۱۵.۸% and ۷.۹% in control group respectivelyand there was a significant association between TLR۷ rs۱۷۹۰۰۸ SNP (P= ۰.۰۰۱, OR = ۳.۳۰۳,۹۵%CI: ۱.۵۸۰-۶.۹۰۶) and the susceptibility of COVID-۱۹ disease. Also, in TLR۷ rs۱۷۹۰۰۹ TT,TC and CC genotypes distribution were ۴۵.۱%, ۴۱% and ۱۳.۹% in case group and ۷۶.۹%, ۱۰.۳%and ۱۲.۸% in control respectively and significant association between TLR۷ rs۱۷۹۰۰۹ SNP (P<۰.۰۰۰۱, OR = ۶.۸۱۸, ۹۵%CI: ۳.۱۰۷-۱۴.۹۶۱) and the susceptibility of COVID-۱۹ disease was foundbetween case and control groups.Conclusion : Our results shows that single nucleotide polymorphisms in TLR۷ (rs۱۷۹۰۰۹) Forboth females and males and TLR۷ (rs۱۷۹۰۰۸) for male gender are considered as a host geneticfactor that could be influenced individual susceptibility to COVID-۱۹ disease.
