Background and Aim : The novel coronavirus disease ۲۰۱۹ (COVID-۱۹) has emerged, hasrapidly extended globally within a short period. Has posed a global threat. Thus, evolve rapid andreliable diagnostic testing. is crucial to control the spread rate of the virus. Reverse transcriptionloop-mediated isothermal amplification (RT-LAMP) as one of these novel approaches to fasterand cheaper testing. The LAMP technique is an alternative to the conventional quantitative RTPCRmethods that do not require expensive instruments and LAMP may achieve higher sensitivityon crude clinical samples than RT-PCR. Using saliva as testing specimen,. LAMP has taken animportant place for the diagnosis of SARS-CoV-۲.Methods : : in this study, we aim to validate the clinical performance of the reverse transcriptionloop-mediated isothermal amplification (RT-LAMP) method for COVID-۱۹ diagnosis, usingRNA was extracted from ۱۰۰ isolated of suspected patients with COVID-۱۹ in selected hospitalsin Tehran. and RT-PCR assay was performed. At the same time, the RT-LAMP was tested directlyfrom nasopharyngeal and oropharyngeal. without RNA extraction from samples.Results : : of ۵۰ positive samples identified by RT-PCR, ۴۵ positive samples nasopharyngeal andoropharyngeal swabs, was detected by Direct RT-LAMP assay. Compared with RT-PCR, overallsensitivity and specificity of RT-LAMP were (۹۰ %) and (۹۵ %) respectively.Conclusion : RT-LAMP have high diagnostic sensitivity and specificity for field diagnosis ofCOVID-۱۹.has few pipetting steps and requires minimal equipment. In addition, RT-LAMP canbe used as a diagnostic method for COVID-۱۹ as an alternative to RT-qPCR in the acutesymptomatic phase of COVID-۱۹. we also that RT-LAMP method paves a way for a largescreening at public domain and hospitals.
