Background and Aim : Pseudomonas aeruginosa is an opportunistic human pathogen that canlead to nosocomial infections which are in turn life-threatening. The increase in antibioticresistance, at an alarming rate, has resulted in a pressing need for alternative therapeuticapproaches such as phage therapy, which hold promise according to several studies. This studyfeatured the isolation and characterization of vB_PaeS_TUMS_P۸۱, a new lytic Pseudomonasphage.Methods : Untreated municipal and hospital wastewater samples were collected and screened. Thehost range determination was performed against standard strains and clinical isolates. Themorphological characterization of the phage was made by transmission electronic microscopy(TEM). Then, the phage genomic DNA was extracted, sequenced and and analysed.Results : Morphologic, genomic and phylogenetic analysis indicated that P۸۱ is a member of thegenus Litunavirus, belonging to Schitoviridae family. The whole-genome sequencing indicatedthat it has a genome of ۷۳,۱۶۷ bp containing ۹۳ predicted coding sequences and genes involved invirulence or lysogeny pathway was nowhere to be found in the genome.Conclusion : The current study indicated that P۸۱ is potentially safe when it comes to therapeuticapplications and it lays the groundwork for further research on treatment of P. aeruginosainfections.
