Background and Aim : Objective: Human gastrointestinal tract microbiota plays roles inmetabolism and interaction with the host cells through several mechanisms such as outermembrane vesicle (OMVs). This study aims to address the effects of Bacteroides thetaiotaomicron(B. thetaiotaomicron) OMVs and its supernatant on the expression of cdh۱,cdh۲,snail۱,zeb۱,andzeb۲ genes involved in Epithelial-Mesenchymal Transition in HCT-۱۱۶ Cell line.Methods : The supernatant was extracted using centrifugation. OMVs extraction was done usingtris-ethylenediaminetetraacetic acid (EDTA)-sodium deoxycholate buffer treatment andcentrifugation. protein content and profile of the OMVs were assessed by sodium dodecylsulphate-polyacrylamide gel electrophoresis (SDS- PAGE) and nanodrop. Human colon cancer(HCT-۱۱۶) cell line as the model of intestinal cancer cells was used to study the effects of B.thetaiotaomicron’s OMVs and supernatant.Results : B. thetaiotaomicron’s OMVs and supernatant increased the expression of snail۱ and cdh۱genes while reduced cdh۲ and zeb۲ expression. It may be able to act as an influential factor inEMT. B.th and its products did not have considerable effect on gene expression of zeb۱.Conclusion : While B. thetaiotaomicron OMVs showed not cytotoxic effects on human intestinalcells, their effects on cdh۱,cdh۲,snail۱,zeb۱, and zeb۲ genes. Based on the features and benefitsthat have been shown in various studies, with more scrutiny they could be considered as promisingfactors and new candidates for treatment. However, more research is needed to examine theireffects in more detail.
