Background and Aim : Antibiotic resistance decreases the eradication rate of Helicobacter pylori(H. pylori). Biofilm formation on the surface of gastric mucosa is a reason to surpass eradicationtherapy. Most of the clinical isolates carry spoT gene which is considered as the major gene ofbiofilm formation in this pathogen. So, this research was designed to study presence of spoT genein H. pylori isolates, and their association with antibiotic resistance and biofilm formation.Methods : Microplate method was used for obtaining MICs of amoxicillin, and clarithromycin for۷۲ isolates of H. pylori. Also, the ability of H. pylori strains for forming biofilm was analyzed.The presence of spoT gene was detected by PCR method and their association with biofilmformation and antibiotic resistance was investigated.Results : The spoT gene were detected in ۶۶(۹۱.۶%) of ۷۲ isolates, respectively. Out of ۶۶ spoTpositiveisolates, ۵۲ were able to form biofilm and none of ۶ spoT- negative isolates were able toform biofilm. The frequency of resistance to amoxicillin, and clarithromycin in spoT+ H. pyloriwere, ۲۴ (۳۶%),and ۴۴ (۶۶.۶۶%), respectively.Conclusion : The study showed that resistance of H. pylori to conventional antibiotics is high inthis region. Hence, there is need for antibiotic susceptibility testing in order to select drugregimens. Additionally, biofilm production and antibiotic resistance of H. pylori was associatedwith harboring of spoT gene , so could be served as a therapeutic target and genetic markers ofantibiotic resistance.
