Investigation of the antibacterial effect of IDR-۱۰۱۸ peptideand chitosan nanoparticles on resistant Pseudomonas aeruginosa isolatedfrom patients with urinary tract infections

Background and Aim : Pseudomonas aeruginosa strains are among the most common causativeagents of urinary tract infections in the hospitals. The excessive use of antibiotics has significantlyincreased resistance to antibiotics in the treatment of UTIs caused by Pseudomonas aeuroginosa.Peptide IDR۱۰۱۸ and chitosan nanoparticles (CNs) have antimicrobial and anti-biofilm activityagainst bacteria. Therefore, in the present study, antibacterial efficacy of chitosan nanoparticlesand peptide IDR-۱۰۱۸ was evaluated against collected P. aeruginosa isolates.Methods : In this study, ۲۰ resistant and ۲۰ sensitive P. aeruginosa isolates to imipenem werecollected from patients with urinary tract infections. The ability of biofilm formation and thepresence of three genes involved in biofilm production including algD, pslD and pelF in theisolates were determined by crystal violet and PCR methods, respectively. The anti-bacterial effectof imipenem, antimicrobial peptide IDR-۱۰۱۸ and chitosan nanoparticles was evaluated bydetermining the MIC by microdilution broth method.Results : The ability of biofilm formation in the isolates was weak (۱۰%), moderate (۳۵%) andstrong (۵۵%). The frequency of presence of three genes related to biofilm formation includingalgD, pslD and pelF was ۹۰%, ۸۰% and ۷۵%, respectively. MIC values for the isolates were ۱۶-۱۶۸ μg/ml for imipenem, ۴۰ and ۸۰ μg/ml for peptide IDR-۱۰۱۸, ۷۵۰ and ۳۷۵ μg/ml for chitosannanoparticles, respectively.Conclusion : Finding alternative approaches such as antimicrobial peptides and nanoparticles canbe effective in treatment of P. aeruginosa infections. The results of our study showed that peptideIDR-۱۰۱۸ and chitosan nanoparticles alone or in combination with imipenem can be consideredas a treatment strategy.
