Background and Aim : Staphylococcus aureus and Methicillin Resistant Staphylococcus aureus(MRSA) are two major causes of nosocomial and community-acquired infections and represent asignificant burden on the healthcare system. The formation of a biofilm decreases the susceptibilityto antimicrobials and immune defenses, making these infections difficult to eradicate. The goal ofthis study was to evaluate the effect of clindamycin eluting liposome nanoparticles, phycocyanineluting liposome nanoparticles, and clindamycin and phycocyanin eluting liposome nanoparticlesagainst biofilm of MRSA and S. aureus isolates.Methods : The fifty clinical isolates of MRSA and S.aureus were collected from patients withbedsores at Loghman Hospital of Tehran-Iran. The biochemical and molecular tests were used todetect S. aureus and MRSA. Liposomes were synthesized using the thin film hydration methodand were characterized by X-ray diffraction, Zeta potential measurement, SEM, and TEM. Theencapsulation of clindamycin and phycocyanin was determined. The effect of the clindamycineluting liposomes, phycocyanin eluting liposomes, and clindamycin and phycocyanin elutingliposomes was evaluated against one, three, and five-day-old biofilm of MRSA and S.aureusisolates using TTC method and then evaluated with Confocal laser scanning microscopy (CLSM).Results : The round-shaped nanoparticles of clindamycin and phycocyanin eluting liposome were۲۳۰ nm and had -۲۲ zeta potential. The encapsulation of clindamycin and phycocyanin in theliposome was ۹۸% and ۸۹%, respectively. Clindamycin and phycocyanin eluting liposomesremoved one, three, and, five-day old biofilms of S. aureus and MRSA at the concentrations of۱۲۵ μg/mL, ۲۵۰ μg/mL, and ۲۵۰ μg/mL, respectively. The vesicular structure of liposomes seemsto enhance antimicrobial activity by improving drug performance compared to free drugs andeliminating S. aureus biofilm.Conclusion : Nanoparticles of antibiotics eluting liposomes can be promising for the treatment ofMRSA and S. aureus biofilms related infections. The phycocyanin pigment with antibiotics elutingliposome can duplicate the effect of antibiotics eluting liposome.
