Molecular identification of amino acid alterations in the OprDporin in clinical isolates of carbapenem-resistant Pseudomonasaeruginosa

Background and Aim : Among several antibiotic resistance mechanisms, mutations in the oprDgene may play an important role in the emergence of carbapenem-resistant Pseudomonasaeruginosa (P. aeruginosa) strains. Therefore, the current study aimed to identification of aminoacid alterations in the OprD porin in clinical isolates of carbapenem-resistant P. aeruginosacollected from patients referred to Ardabil hospitals.Methods : The presence of the oprD gene was detected using polymerase chain reaction (PCR)technique in carbapenem-resistant P. aeruginosa strains and then five strains positive for oprDgene were selected randomly and sent for DNA sequencing. The results were compared with thereference strain PAO۱.Results : As shown in Figure ۱, the most frequent amino acid alterations in the OprD porin were۲۱۰ A→I, ۲۰۲ Q→E, ۱۹۰ A→K, ۱۸۶ A→P, ۱۷۰ L→F, ۱۱۵ T→K and ۱۰۳ S→T.Conclusion : Our results revealed that mutations in the oprD gene play are involved in theemergence of carbapenem-resistant P. aeruginosa strains in Ardabil city hospitals.
