Inhibitory effect of prodigiosin pigment produced by Serratiamarsecsenes on Trichomonas vaginalis

چکیده مقاله:

Background and Aim : Microbial pigments are a type of secondary metabolite that can play agood role due to its antimicrobial properties. Prodigiosin is an antimicrobial tri-pyrrole pigmentthat can be extracted from Serratia marcescens . Trichomonas vaginalis is a protozoan that entersthe human reproductive system and causes trichomoniasis. Trichomoniasis is the most commonnon-viral sexually transmitted disease treated with metronidazole. Side effects and drug-resistantare the two main problems of this drug that make doctors look for new treatments. This studyaimed to extract prodigiosin pigment from Serratia marcesens to evaluate the antiparasiticproperties of this pigment against Trichomonas vaginalis in vitro.Methods : For this study, Serratia marcescens strain RTCC ۲۲۸۱ was purchased from RaziInstitute and mass cultivated. The pigment prodigiosin was extracted using Ethyl acetate andacetone tools and the relativity of the pigment was confirmed by TLC chromatography, UV-Visspectrophotometr, and FTIR and its effect on Trichomonas vaginalis and PC۱۲ cell line wasexamined in different concentration.Results : Prodigosin pigment at ۱۰۰۰۰ μg / ml in ۲۴ hours and ۵۰۰۰ μg /ml and ۲۵۰۰ in ۴۸ hourscaused ۱۰۰%, ۹۲.۸۹% and ۸۴.۲۶% growth inhibiton of the parasite. Its IC۵۰ in ۴۸ hours ۱۱۲.۸ μg/ ml and the CC۵۰ of this pigment on the PC۱۲ cell line reached ۸۴.۴ μg / ml. All test steps wereperformed with three replications and the statistical results were calculated using Graphpad Prismsoftware version ۹.Conclusion : According to the results, this pigment was tested on Trichomonas vaginalis and itsselective index on the cell line was ۷.۴۶ times (SI = ۷.۴۶), so it can be concluded that prodigiosinpigment had an antiparasitic effect and More And more comprehensive study of prodigiosincomposition in vitro and in vivo is suggested.
