Antiparasitic activity of pyocyanin pigment produced byPseudomonas aeroginosa against Trichomonas vaginalis

چکیده مقاله:

Background and Aim : Pyocyanin pigment is a blue redox compound that is metabolically activeand has therapeutic effects on eukaryotic and prokaryotic cells. The opportunistic pathogenPseudomonas aeruginosa is the only non-fermenting lactose gram-negative bacterium thatproduces the pigment pyocyanin. Trichomonas vaginalis is a flagellar protozoan that causestrichomoniasis, the most important non-viral sexually transmitted disease in the world. The maintreatment for this infection is metronidazole, which scientists are looking for new ways to treatdue to the observed side effects and drug resistance. This study aimed to investigate the effect ofpyocyanin pigment extracted from Pseudomonas aeruginosa on Trichomonas vaginalis and alsothe effect of pyocyanin toxicity on the PC۱۲ cell line.Methods : For this purpose, Pseudomonas aeruginosa RTCC۱۴۷۴ strain was purchased from RaziVaccine and Serum Research Institute and pyocyanin pigment was extracted from this bacteriumwith the help of chloroform. The relative purity of the pigment was confirmed by thin layerchromatography, spectrometry UV-Vis and FTIR and We examined its effect on differentconcentrations against Trichomonas vaginalis parasite and PC۱۲ cell line.Results : Pyocyanin pigment at concentrations of ۱۰۰۰۰μg / ml in ۲۴ hours and concentrations of۵۰۰۰ and ۲۵۰۰ μg / ml in ۴۸ hours inhibited parasite growth ۱۰۰%. Its IC۵۰ level in ۴۸ hours was۱۷.۴۴ μg / ml and the CC۵۰ of this pigment was obtained on the cell line at ۹۳۰ μg / ml. Allexperimental steps were performed with three replications and statistical results were obtainedwith the help of GraphPad Prism software version ۹.Conclusion : According to the results, this pigment was effective against Trichomonas vaginalisparasite, and its selective index on cell line was ۵۳ times (SI = ۵۳.۳۲), so more and morecomprehensive studies to investigate the composition of pyocyanin in vitro and in vivo aresuggested.
