Inhibitory effect of clove plant extract on Staphylococcusaureus bacteria in vitro and its comparison with selected antibiotics

Background and Aim : Nowadays due to the increasing incidence of antibiotic resistance,theneed to find new and safe antibacterial compounds has become one of the main priorities ofmedical researches. Staphylococcus aureus is a vital bacterial pathogen that is considered as oneof the significant causes of nosocomial infections. Antibiotics in this bacterium and the emergenceof resistant strain of vancomycin are now a major hospital challenge in the field of infectioncontrol. Today, the use of plants in the treatment of resistant infections is considered as anantibacterial source. In this study, the inhibitory effect of clove on the standard strain ofStaphylococcus aureus in vitro by disk diffusion method was investigated and the results werecompared with standard therapeutic antibioticsMethods : In this study, Staphylococcus aureus was cultured on Mueller Hinton agar (MHA) andincubated for ۲۴ hours, additionally ۰.۵ McFarland suspension was prepared. Afterwards, grindingthe clove plant an aquatic extract was prepared from the clove powder. In order to determine thesensitivity, ۳ wells were created in each culture medium and different extracts were administeredseparately from ۱۰,۱۵,۲۵,۵۰,۷۵ to ۱۰۰ μl, respectively. It was poured into the wells andsimultaneously other selective antibiotics of Vancomycin, Chloramphenicol, Tetracycline wereplaced in other plates. After measuring the created growth inhibition halos, the obtained resultswere evaluated and the diameter of growth inhibition halos was measured carefully.Results : The outcomes of this experiment showed that the halos created by the clove herb aresurprisingly effective and in some volumes are equal to the selected antibiotics. On the other hand,the more extract’s concentrations were increased, the larger inhibition halos were created.Subsequently, the results of this study showed that the largest growth inhibition halo was relatedto the volume of ۷۵ and ۱۰۰ ?l with diameter of ۲۹ and ۳۰ mm and the smallest ۲۰ mm with avolume of ۱۰ ?l were measured. The halo of non-growth of bacteria in the presence of vancomycinand tetracycline antibiotics was reported to be ۲۴ and ۲۷ mm, respectively.Conclusion : The results indicate that the aqueous extract of clove plant is strongly effective inpreventing the growth of Staphylococcus aureus and can be used as a plant alternative to selectedantibiotics in the treatment of infections caused by Staphylococcus aureus
