New treatment of bacterial infections caused by Escherichiacoli based on exosomes secreted from stem cells

چکیده مقاله:

Background and Aim : Exosomes derived from MSCs are used to detect and treat bacterial andviral infections. Exosomes help in the healing process by delivering miRNA and mRNA, peptides,proteins, cytokines, and lipids, as well as communicating with other organs in the body. Theadvantages of these Extracellular vesicles (EVs) cells are their small size, high stability, lowfunction, repair of tissue damage caused by infection, and reduced inflammatory factors. Thecurrent study's purpose was to determine the use of Exosomes for treatment. E. coli belongs to theEnterobacteriaceae family, which is known to cause various infectious diseases. We will cover thetwo treatments for disorders induced by exosomes. E. coli is implicated in the formation of liverabscesses, which may lead to mortality in cases with comparable infections and inflammatory solidreactions. AIEC is another strain of E. coli that causes common Crohn's disease. To survive inintestinal epithelial cells, these bacteria enter macrophages and generate inflammation andcytokines. As the initial line of bacterial therapy, we utilized antibiotics.Methods : In both studies, qRT-PCR methods were used to evaluate the number of exosomes andassess the inflammatory response in the infected cells. Then, we purified and secreted Exosomesreleased by T۸۴ cells (Exo-uninfected) and infected human intestinal epithelial and revealedaddition of them.Results : According to the analysis of the combination of imipenem and exosomes derived fromstem cells, there is a significant synergistic effect and a decrease in the secretion of inflammatorycytokines, nitric oxide, and the amount of apoptosis in infected cells. Analyzes indicate thattransmission of these miRNAs was significantly increased from exosomes to recipient cells toinhibiting autophagy-mediated clearance of intracellular AIECConclusion : Due to increased drug resistance, Escherichia coli, which leads to a wide range ofdiseases, the use of exosomes as an effective non-antibiotic treatment method has been applied inthese studies against infections caused by this bacteria there are some limitations to this study thatneed to be acknowledged and considered in future.
