Significance of circulating miR-۱۰۷ in type۲ diabetes mellitus

Backgrounds: MicroRNAs (miRNAs) are involved in various biological processes, such as celldevelopment, proliferation, differentiation, apoptosis and metabolism. It has been shown thatthere is selective expression of circulating miRNA that may correlate with diabetic conditions.Here, we evaluated the expression of circulating miR-۱۰۷ by quantitative PCR in blood frompatients with type ۲ diabetes (T۲D) and healthy controls.Materials and Methods: In this case-control study, miR-۱۷۰ expression was evaluated in ۳۰blood samples from patients with T۲D and ۳۰ controls. Total RNA was extracted using Trizolreagent. The integrity and quality of RNA was evaluated gel electrophoresis andspectrophotometry. Quantitative real‐time polymerase chain reaction (qRT-PCR) was performedfollowing synthesis of complementary DNA (cDNA). Specific primers were designed using theOligo ۷ software and were subsequently synthesized. Each run of qRT-PCR was completed witha melting curve analysis. Quantitative measurements were performed in triplicate and relativeexpression was measured using comparative Ct method (۲-ΔΔCt) and normalized to U۶ snRNA asinternal control. Statistical analyses were performed using GraphPad Prism ۸.۰. P < ۰.۰۵ wasconsidered statistically significant.Results: A single peak was observed in all the dissociation curves and a single band was seen onthe gel at the correct product size for each gene. miR-۱۰۷ expression in patients was significantlyhigher than that in the controls (۱.۹۴ ± ۰.۰۴ vs۱.۰۱ ± ۰.۰۲ p < ۰.۰۰۰۱).Conclusion: According to the data of this study, miR-۱۰۷ expression level appear to be effectivein the emergence of T۲DM.
