Backgrounds: Colorectal cancer (CRC) is the third most common type of cancer in the worlddue to the lack of noninvasive and accessible prognostic and diagnostic biomarkers for earlydetection of this disease. Long noncoding RNAs (LncRNAs) are of paramount importance in theunderlying molecular mechanisms of CRC initiation and progression, and can be as a potentialbiomarker in it. According to the previous studies, MTX۲ acts as a tumor suppressor by bindingto miR-۵۷۴-۵p and increase SMAD۴ expression in esophageal squamous cell carcinoma,cervical and stomach cancers. On the other hand, decrease of SMAD۴ expression is associatedwith CRC metastasis and chemotherapy resistant. For these reasons, the aim of this study wasinvestigation of MTX۲ expression in CRC tissue samples and evaluation of its potential asdiagnostic biomarker in this cancer for first time.Materials and Methods: The colorectal tumor samples and adjacent normal samples werecollected from thirty patients. After RNA extraction and cDNA synthesis, Real-time PCR wasused to measure the LincMTX۲ expression.Results: The qPCR results showed the expression of LincMTX۲ was down-regulated in thirtypaired colorectal cancer specimens (P-value = ۰.۰۰۰۱). The AUC of ROC curve was ۰.۷۳۰۵ andrevealed that the expression level of LincMTX۲ can detect up to ۷۳% of cases of colorectalcancer.Conclusion: The considerably down-regulation of LincMTX۲ in CRC tumor samples and itspotential as biomarker highlight the need to further investigation of this LncRNA in largerpopulation and cohort studies.
