Backgrounds: GBM is the most aggressive diffuse glioma of astrocytic lineage. GBM is still anincurable disease with a ۱۵-month median survival rate. The purpose of this study was toevaluate alternations in Akt gene expression in the Glioblastoma T۹۸G cell line under treatmentwith thiosemicarbazones complexes (Ni).Materials and Methods: Ni-thiosemicarbazones complexes were prepared in ۵۷ and ۶۱μM. TheT۹۸G cell line, was purchased from Pasteur Institute of Iran at passage ۱, and then treated by Nithiosemicarbazonescomplexes in two groups after cell passage in ۴۸h. Then RNA extraction andcDNA synthesis were performed and the expression of Akt gene and GAPDH were evaluated byReal-Time PCR. Finally, the results of Real-Time PCR were analyzed.Results: Evidence showed that after treatment of the cell line with thiosemicarbazonescomplexes of Ni, Akt gene decreased in both concentrations of ۵۷ and ۶۱ μM in ۴۸ hours, whichwas statistically significant. The rate of reduction in Akt gene expression treated withthiosemicarbazones complexes of Ni at concentrations of ۵۷ and ۶۱ μM was ۰.۰۰۱ and ۰,respectively.Conclusion: The results of T۹۸G cell line treatment with Ni-thiosemicarbazones complexes andcomparison with GPDH control gene showed that Ni- thiosemicarbazones complexes can beeffective in reducing the expression of Akt gene as an oncogene. Changes in Akt gene expressionshowed that with increasing concentration, the greatest reduction in expression was observed, sothe optimal effect of the drug depended on its concentration.
