The effect of alpha lipoic acid on cell survival and fis۱ gene expression in a Parkinson's disease cell model

Backgrounds: Mitochondria are known as semi-independent and dynamic organelles. Rotenonewas used as an inhibitor of mitochondrial complex I to induce the Parkinson's disease model. Inthe present study, to further understand the role of mitochondrial dynamics in PD, it wasdetermined to investigate the effective rotenone concentrations in SH-SY۵Y neurons viability aswell as the effect of alpha lipoic acid on cell survival in stress condition induce by rotenone.Genomic expression level of fis۱ gene, in SH-SY۵Y cell line treated with rotenone and alphalipoicacid was evaluated.Materials and Methods: The effect of rotenone toxicity on SH-SY۵Y cell growth in presence ofrotenone and alpha-lipoic acid was evaluated by MTT method. The expression of fis۱ gene wasthen evaluated by real-time PCR.Results: Based on IC۵۰ calculations, the toxicity of rotenone was determined at concentrationshigher than ۱ μM. Toxicity effect of two concentrations of ۲.۵- and ۵- μM rotenone for ۲۴ and۴۸ hours’ treatment showed a significant decrease in SH-SY۵Y cells survival (P ˂ ۰.۰۰۰۱).Improved cell viability was observed in presence of ۲۰ μM alpha-lipoic acid in SH-SY۵Y cellstreated by rotenone (P = ۰.۰۴). The qPCR results of fis۱ gene expression indicated a significantincrease in SH-SY۵Y cells treated with ۵ μM rotenone (P < ۰.۰۰۲).Conclusion: The results of this study show that the expression of a mitochondrial fission gene isaltered in rotenone-treated SH-SY۵Y cells in culture medium. Thus, mitochondrial dynamicsmay involve in the pathogenesis or control of Parkinson's disease.
