Differential gene expressions of CALM۱, PSMD۶, and AK۱۲۴۷۴۲ long non-coding RNA in Cumulus cells from PCOS patients vs. normal control women

Backgrounds: One of the well-known causes of subfertility is polycystic ovary syndrome(PCOS). Genetic components play a critical role in the etiology of PCOS. The recognition ofdifferentially expressed genes in PCOS patients might provide a better understanding of thepathophysiology of this syndrome and paves the way for novel therapeutics. Gene expressionprofiles in cumulus cells could be used as biological criteria for embryo competence and theiranalysis might lead to important molecular information about embryo quality. CALM۱, PSMD۶,and AK۱۲۴۷۴۲ are three well-known genes associated with embryo development. Therefore, theobjective of this study was to compare the expression of CALM۱, PSMD۶, and AK۱۲۴۷۴۲ genesin the cumulus cells of infertile PCOS patients with their expression in the cumulus cells of thedonor fertile group.Materials and Methods: Cumulus cells were collected from the follicular fluid of ۳۳ patientswith PCOS as the experimental group and ۳۳ cumulus donor women who referred to theinfertility center for egg donation as the control group. Cumulus cells were frozen until genetictesting. The expression of CALM۱, PSMD۶, and AK۱۲۴۷۴۲ genes was detected by real-timePCR.Results: CALM۱ and AK۱۲۴۷۴۲ gene expressions significantly increased (CALM۱ P = ۰.۰۰۳)(AK۱۲۴۷۴۲ P = ۰.۰۰۰) and PSMD۶ expression significantly decreased (P = ۰.۰۰۲) in the PCOSgroup compared to the cumulus donor group.Conclusion: Therefore, our findings suggested that may be the impact of PCOS on fertility isbecause of the changes in the expression levels of genes affecting the reproductive processincluding CALM۱, AK۱۲۴۷۴۲, and PSMD۶.
