Background: Breast cancer (BC) is one of the most common cancers among women around theworld as well as in Iran. Non-coding RNAs play a key role in the development of BC, their useas biomarkers in diagnosis and prognosis is becoming increasingly useful. In recent years, theypay more attention to circularRNA(circRNA) due to its high stability and the possibility of earlydetection moreover the discovery of their roles in the pathogenesis of the disease, and their hightherapeutic and diagnostic potential have opened new horizons for research.Materials and Methods: ۲۹ BC tumors and their adjacent normal tissues were collected fromMotamed institute of breast cancer research (Tehran, Iran). The samples were snap-frozen andstored at −۸۰ ◦C. Before sample collection, circHIPK۳(has-circ-۰۰۰۰۲۸۴) expression in breastcancer tissues was analyzed using qRT-PCR. CircInteractome(https://circinteractome.nia.nih.gov) was performed to detect circRNA/miRNA targetingrelationship. Additionally, potential miRNAs that can be targeted by circHIPK۳ were predictedby the bioinformatics method. miRTargetlink database (https://ccb-web.cs.unisaarland.de/mirtargetlink) and DIANA database (http://diana.imis.athenainnovation.gr/DianaTools/) were used to predict interactions of miRNAs with circHIPK۳.circHIPK۳ has carcinogenic or anti-tumor effects on different cancers.Results: The lower expression levels of circHIPK۳ in Her۲+ BC tissues were shown comparedto paired adjacent normal tissues and Her۲- tissues with P-value (P <۰.۰۰۵۴). In addition, ۱۰miRNAs can be targeted by candidate circRNA, which was predicted based on bioinformaticsdatabases. Moreover, miRNAs selection tools predicted that miR-۱۹۲, miR-۳۲۶, and miR-۳۶۵can be targeted by circHIPK۳.Conclusion: Our findings revealed that circHIPK۳ facilitates BC development and progression,providing a novel therapeutic target for BC and could serve as a prognostic biomarker of BC.
