The study of miR-۱۵a and BACE۱ gene expression in patients with Alzheimer's disease

Backgrounds: Alzheimer’s disease (AD), the most common form of dementia is aneurodegenerative disorder. BACE۱ plays a critical role in AD pathophysiology by generation ofAβ which leads to accumulation of extracellular Aβ plaques. Some miRNAs like miR-۱۵a aredysregulated in AD patients. Furthermore, BACE۱ is a target gene of some miRNAs. Theobjective of this study was to investigate the relative expression level of BACE۱ and miR-۱۵agenes in Alzheimer patients.Materials and Methods: Total RNA isolated from whole blood samples were used to synthesizecDNA for quantitative real time polymerase chain reaction analysis. The relative expression ofBACE۱ and miR-۱۵a were measured using comparative Ct method (۲ −ΔΔCt) and normalized toGAPDH and U۶ as internal controls. Statistical differences were analyzed by GraphPad Prism (v.۸.۰) statistical software or Student’s t-test.Results: The results showed that BACE۱ was highly expressed in patients as compared tocontrols and this difference was statistically significant. There was also a significant decrease inmiR-۱۵a expression in patients’ blood (۰.۷۲ ± ۰.۰۸) as compared to control subjects (۰.۹۵ ±۰.۰۹).Conclusion: According to our funding so far, a statistically significant increase in BACE۱, and adecrease in miR-۱۵a expression exist in AD samples. This suggests that an interaction/relationbetween these two genes may exist in AD patients. Our finding is in line with the previousreports showing the miR-۱۵a expression was altered in AD patients so that both the down andup-regulations were reported in different studies. Furthermore, studies are needed to reveal thealternative underling pathway by which these genes interfere to lead to the AD pathology.
