چکیده مقاله:
Backgrounds: BMSCs can differentiate into neuron- like cells in vitro conditions. Importantgenes in this process are ELP۱, PHOX۲B and MAP۲. Glabridin derived from the roots oflicorice. Use of Glabridin on cell differentiation was studied. Changes made in the expression ofELP۱, PHOX۲B and MAP۲ genes by Glabridin effect in the pathway of differentiation ofBMSCs to neuron- like cells was studied in this article.Materials and Methods: BMSCs was incubated. Toxication levels of Glabridin weredetermined by MTT Assay test in concentrations of ۵, ۱۰, ۲۰, ۴۰ and ۸۰ μM. CDNA wassynthesized and the amount of change in gene expression was studied by real time PCR.Results: Glabridin fatality dosage was tested by MTT in ۴۰ and ۸۰ μM concentrations and basedon ۲۴-hour analysis, ELP۱, PHOX۲B and MAP۲ gene expression in samples incubated withGlabridin had significant increase compared to the control group. Conclusion: Results showed that Glabridin of BMSCs cause increased ELP۱, PHOX۲B andMAP۲ gene expression resulting in increase in their differentiation into neuron-like cells.
