Protective effect of HLA-E*۰۱۰۱/*۰۱۰۳ genotype in survival of patients after allogeneic hematopoietic stem cell transplant

چکیده مقاله:

Backgrounds: HLA-E is located on the non-classical major histocompatibility complex class Iand acts as the ligand for natural killer cells. Consequently, it has a main role in the regulation ofinnate immune responses by involving cell identification by natural killer cells. Differences inexpression levels among HLA-E alleles have been suggested to affect transplant outcomes. Inthis study, we evaluated the effects of different HLA-E genotypes on allogeneic hematopoieticstem cell transplant in southern Iran.Materials and Methods: We investigated ۲۰۰ patients who underwent allogeneic hematopoieticstem-cell transplant and ۱۰۰ normal participants (control group) in a case-control study.Detection of HLA-E polymorphisms was performed using a sequence-specific primerpolymerase chain reaction method.Results: Statistical analyses indicated that genotypes in the transplant group were not distributedin accordance with Hardy-Weinberg equilibrium (χ۲ = ۷۶.۵۶; P < .۰۰۱), whereas genotypes inthe control group were distributed in accordance with Hardy-Weinberg equilibrium (χ۲ =۰.۳۹; P = .۵۳). No significant differences were observed in cumulative incidence of acute (P =.۷۶; hazard ratio = ۰.۸۰; ۹۵% confidence interval, ۰.۱۹-۳.۳۱) and chronic (P = .۷۵, hazard ratio =۰.۰۴۸; ۹۵% confidence interval, ۰.۰۰).Conclusion: Genotypes of the HLA-E molecule did not correlate with acute and chronic graftversus-host disease in hematopoietic stem cell transplant recipients except for the HLAE*۰۱۰۱*/*۰۱۰۳ genotype, which was protective in survival of our study patients.
