Study of NGF, PAX۳, and NSE genes expression in bone marrow-derived mesenchymal stem cells after treatment by Glabridin for differentiation into nerve-like cell

چکیده مقاله:

Backgrounds: Glabridin extracted from the licorice root. The potential differentiation ofBMSCs into nerve-like cells has been approved in the in-vitro, the effectiveness of Glabridin indifferentiation of these cells by the mediating role of gene expression to find a solution forimproving the nervous system disorders. Changes the expression of the genes NGF, PAX۳, NSEin differentiation of BMSCs into the nerve cells by Glabridin were studied.Materials and Methods: BMSCs with different Glabridin concentrations of ۵, ۱۰, ۲۰, ۴۰, ۸۰μM were induced for ۲۴ hours. The glabridin toxicity was tested by MTT assay. The changes inthe expression of NGF, PAX۳, NSE genes were evaluated by Real-time PCR.Results: MTT showed that the cells died in ۴۰, ۸۰ μM concentration in ۲۴h treatments, theexpression of the genes NGF, NSE increased compared to the control group, the expression ofgene PAX۳ decreased.Conclusion: Treatment of BMSCs with Glabridin increased the expression of NGF, NSE genesand decreased the expression of PAX۳ to these cells into neuron-like cells.
