Backgrounds: In the present paper, we effort to inquire the effects of alcoholic extract Silybummarianum L. (AESM) on in vitro and in vivo anti-inflammation and the alleviation of cartilagedegradation in the Rabbit model of monosodium iodoacetate (MIA)-induced osteoarthriti. Werealize that Silybum marianum L can efficiently and dose-dependently repress the expression ofpro-inflammatory cytokines mRNA, including iL-۶, iL-۱α, iL-۱۸, and TNF-α in LPS-stimulatedsynoviocytes. Researchers indicate that Silymarin is a compound contains various properties likeanti-inflammatory hepatoprotective, antioxidant, heart-protective, hypocholesterolemic, antidiabetic,anticancer, and cardio-protective activities. Clinical studies have been demonstratedthat Silymarin has very rare side effects at high doses (>۱۵۰۰ mg/day).Materials and Methods: RNA extraction by ®TRIzol method (SinaClon, Iran), Convert RNAto cDNA (Malaysia‐Selangor), evaluation of gene expression such as iL-۶, iL-۱α, iL-۱۸, andTNF-α by RT-PCR, simulation of OA with the help of MIA, extraction with a rotary evaporatorvacuum device, the MTT technique, isolation and culture of RFLS.Results: AESM decreased the expression of iL-۶, iL-۱α, iL-۱۸, and TNF-α genes in RFLS cellsand in cartilage.Conclusion: AESM can compete significantly with drugs such as dexamethasone and ibuprofenin the treatment of OA. Our experiments indicated that consumption administration of AESMreduces the expression of TNF-α, iL-۶, iL-۱α and iL-۱۸ genes and can compete well withcommon drugs in the treatment of OA.
