The anti-proliferative effect of Moringa olifera aqueous leaf extracts on acute lymphoblastic leukemia cells

Backgrounds: Acute lymphoblastic leukemia (ALL) is the most common hematologymalignancy in children. Use of natural compounds in the treatment of ALL has been considered,due to the side effects and toxicity of traditional chemotherapy. The aim of this study was toevaluate the anti-proliferative effect of Moringa olifera aqueous leaf extract in Raji and Jurkatacute lymphoblastic cell lines.Materials and Methods: After preparing the aqueous leaf extract by soaking, the cells werecultured separately and treated with different concentrations of the extract for ۴۸ hours. Cellviability and growth inhibition were assessed by trypan blue and MTT assay. Changes in P۲۱proapoptotic gene expression were assessed by real time PCR. Data were analyzed using SPSS۲۳ and one-way ANOVA method.Results: Concentrations of ۵۰ to ۲۵۰ μg/ml of the extract significantly reduced the growth ofRaji and Jurkat cell lines (p<۰.۰۰۱). The highest growth inhibition rates for concentration ۵۰μg/ml were ۷۳.۴% for Jurkat and ۷۸.۵% for Raji cell lines. IC۵۰ was obtained at a concentrationof ۱۵۰ μg/ml. Treatment of cells with this concentration resulted in a ۱۴۷-fold increase in P۲۱gene expression for Jurkat (p = ۰.۰۰۹۱) and ۱۵-fold increase for Raji (p = ۰.۰۰۲۶) cell lines.Conclusion: Moringa olifera aqueous leaf extracts appeared to be able to inhibit growth andincrease apoptosis in Raji and Jurkat cell lines. It can be concluded that the use of Moringaleaves as a medicinal or nutritional supplement is useful in the treatment of ALL.
