Backgrounds: Diabetes Mellitus (DM) is a heterogeneous metabolic disorder that let tohyperglycemia and further to different complications such as retinopathy, nephropathy, delayedwound healing, and. Despite many studies has been conducted on diabetic neuropathy, diseasedevelopment and drug discovery investigations are yet essential at preclinical stage usingappropriate animal models. Zebrafish (Danio rerio) because of its features such as transparencyat the early developmental stages, easy husbandry and care, regenerative capacity for mostorgans, along with extremely fast development and similarity in genomic and biologicalpathways with human in different diseases including diabetes, is an ideal disease modelorganism.Materials and Methods: In this study, immersing method was used to induce hyperglycemiaand investigate its influence on central nervous system. To induce DM in Zebrafish, adulttransgenic Zebrafish (mbp:(eGFP-NTR)) that was generated by Tol۲ transposase technique,immersed in ۲۲۰ mM glucose solution for ۳۰ days. In order to measure blood glucose level andevaluate the stability of hyperglycemia and other biochemical parameters for subsequent studies,blood collection performed repeatedly.Results: All experiment conducted in triplicate and results showed that the average bloodglucose level in the treatment group (۱۳۷mg/dl) increased by ۴ times compared to the controlgroup (۳۸mg/dl).Conclusion: These data suggested that the exposure concentration and time are enough to inducehyperglycemia and impaired peripheral glucose metabolism in adult zebrafish, and thistransgenic model could be used to further study the effects of hyperglycemia on neural system.
