Bioinformatics’ analysis of microRNA in angiogenesis pathway of breast cancer: biomarker role of microRNA

Backgrounds: Breast cancer is commonly invasive neoplasms in women. Various factors play arole in the development and promotion of breast cancer; most of them involve changes in theexpression of certain genes, including those encoding noncoding RNAs. Recently, many studieshave elucidated the key roles of microRNAs (miRNAs) in controlling gene expression duringcancer development. MiRNAs are small subclass of ncRNAs with almost ۱۹–۲۴ nucleotides.Multigene regulatory features of these small RNA molecules allow them to change signalingpathways, facilitate or block signal transmission to downstream effectors in various ways.MiRNAs can regulate key signaling pathways positively or negatively, therefore they can affecttumorigenesis.Materials and Methods: We used NCBI database to nominate a gene called “MMP۹”, which ishighly expressed in breast cancer. We demonstrated that MMP۹ is involved in the developmentof angiogenesis in cancers. By studying in miRTargetLink database we found that ۶ miRNAshave strong interaction with MMP۹ gene. We selected two of them (hsa-miR-۴۹۱-۵p and hsamiR-۲۹b-۳p) for more investigations. Then, we used DIANA TOOLS database and drawingHeatmap by this database we found that selected miRNAs have high expression in PI۳K/AKTsignaling pathway and pathway in cancers.Results: MMP۹ gene has high expression and it has specific roles in pathway in cancer. Otheranalysis showed that our selected miRNAs have high expression in PI۳K/AKT signalingpathway and pathway in cancer. So, these miRNAs can bind to our selected gene and change itsexpression and function that can make some changes in cancer.Conclusion: We established that hsa-miR-۴۹۱-۵p and hsa-miR-۲۹b-۳p interaction with this genecan change important pathways in cancer and they can play significant roles so, they can be usedas a functional biomarker and diagnosis in breast cancer.
