Backgrounds: Cholesterol is a lipid-like substance with the chemical structure of C۲۲H۴۵OHthat is produced in our body and is also found in food becomes. There are different types ofcholesterol inside the cell and in the bloodstream. Excessive increase in blood cholesterol causesvarious diseases, including hypercholesterolemia. Research shows that mutations in certain genesare associated with high cholesterol. One of these genes is the ABCG۸ gene, which is located onchromosome ۲ and transports cholesterol to the bile. The aim of this study was to investigate theassociation of ۲۶۵۰۰۰۰ and ۶۵۴۴۷۱۴ polymorphisms in the ABCG۸ gene with high cholesterol.Materials and Methods: We collected ۹۰ peripheral blood samples and extracted the genomicDNA of each sample. Then, we generated and analyzed the gene using Tetra Arms PCR.Results: ۲۶۵۰۰۰۰ polymorphism genotypes include homozygous GG, TT and heterozygous GTgenotypes. The T allele has been linked to high cholesterol. in the other hand, recessive GGgenotype was observed in ۱۷.۵% of patients and ۴۷.۵% of healthy individuals, which indicates ahigher percentage of this genotype in healthy individuals. Homozygous CC and GG genotypesand heterozygous GC genotypes were also observed in polymorphism ۶۵۴۴۷۱۴. Thepredominant GG genotype was ۵% in patients and ۶۲.۵% in healthy individuals. On the otherhand, the recessive CC genotype was ۴۲.۵% in patients and ۷.۵% in controls. The P-Value for۲۶۵۰,۰۰۰ polymorphisms was less than ۰.۰۵ and equal to ۰.۰۱. This shows that there is asignificant difference between the alleles of healthy and sick people. The value of P-Value forpolymorphism ۶۵۴۴۷۱۴ was also zero.Conclusion: The predominant TT genotype related to polymorphism is ۲۶۵۰۰۰۰ and thehomozygous CC genotype related to polymorphism ۶۵۴۴۷۱۴ is factors for increasing cholesterolin people with high cholesterol.
