Backgrounds: Colorectal cancer (CRC) is the third most common cancer in the world. Recentstudies show that long non-coding RNAs (LncRNAs) play an important role in tumor formation,proliferation, migration, and apoptosis. This study aimed to investigate the expression ofPWRN۱ (Prader-Willi Region Non-Protein Coding RNA ۱) in colorectal cancer. This lncRNAwas selected based on bioinformatics data and there has been no research report on its expressionin tissue samples of CRC at present. Bioinformatics studies have shown that this LncRNA maybe involved in gastric, renal, and colorectal cancers, which are involved by thePTEN/Akt/MDM۲/۵۳p pathway.Materials and Methods: The colorectal tumors along with the corresponding adjacent normaltissues were collected from thirty patients attending Milad Hospital, Isfahan, Iran. All thesamples were collected following the guidelines issued by the Ethics Committee of IsfahanUniversity of Medical Sciences (approval number: ۶۳۰۷۶۰۳). After total RNA extraction fromsamples and cDNA synthesis, Real-time PCR was used to measure the PWRN۱ expression.Results: The qPCR results showed the expression level of PWRN۱ was down-regulated byroughly ۸.۵ times in thirty paired colorectal cancer specimens (p-value = ۰.۰۰۰۴).Conclusion: The statistical analyses demonstrated that PWRN۱ could serve as the diagnosticbiomarkers to distinguish the tumor samples. Upon further validation in the bigger and moresophisticated studies, this lncRNA could have implications in clinics to improve the earlydiagnosis of colorectal cancer.
