چکیده مقاله:

Gene therapy is a new method in modifying genetic diseases. The desiredgene enters directly or indirectly into the target cells. Carriers play animportant role in the process of gene delivery. The two main types of vectorsare viral and non-viral vectors, which are discussed in this article. Viralvectors include retroviruses, adenoviruses, adeno-associated viruses, andlentiviruses and non-viral vectors include liposomes, cationic polymers, andmagnetic nanoparticles. Gene delivery efficiency through viral vectors isreported to be ۱۰ to ۱۰۰۰ times higher than non-viral vectors. In non-viralvectors, immunity is considered as an advantage and low gene transferefficiency as a disadvantage, and in viral vectors, productivity is consideredas an advantage and immunogenicity and pathogenicity as a disadvantage.Research shows that gene expression with viral vectors is far higher thanwith non-viral vectors. Researchers have found that the uptake of DNA intocells with a non-viral vector is better than the viral vector, but in later stepssuch as the release of DNA into the cytoplasm and entry into the nucleus, aviral vector such as adenovirus is better. Gene therapy through non-viralvectors that are used with appropriate protocols with their own strengths andweaknesses seems to be useful for the treatment of some diseases. Also, viralvectors are uniquely suitable for the treatment of genetic diseases.
