The effect of resveratrol on expression of matrix metalloproteinase ۹ and its tissue inhibitors in vascular smooth muscle cells

BACKGROUND: Matrix metalloproteinase ۹ (MMP-۹) is involved in extracellular matrix degradation and remodeling. An increase in MMP-۹ expression by vascular component cells plays an important role in atherosclerotic plaque formation and rupture. Resveratrol, a polyphenolic substance, was suggested to play a role in preventing the progress of atherosclerotic disease. The aim of this study was to investigate the effect of resveratrol on MMP-۹ and tissue inhibitors of metalloproteinases (TIMPs) in vascular smooth muscle cells (VSMCs) after treatment with H۲O۲.METHODS: Cultured VSMCs were pre-treated with ۰.۲ mM of H۲O۲ before stimulation with different concentration of resveratrol. Expression of MMP-۹, TIMP-۱, and TIMP-۳ genes were measured using real-time polymerase chain reaction (PCR) method, and MMP-۹ protein level was detected using western blot analysis.RESULTS: Resveratrol at ۱۲۰ μmol/l concentration reduced the elevated level of MMP-۹ induced by H۲O۲ in VSMCs as ۱.۸۵ ± ۰.۳۵ folds (P < ۰.۰۵) and ۸.۷۰ ± ۱.۲۰ folds (P < ۰.۰۵) after ۲۴ and ۴۸ hours, respectively. Resveratrol increased the diminished level of TIMP-۱ induced by H۲O۲ as ۲.۵ ± ۰.۴۸ folds following the treatment with ۱۲۰ μmol/l after ۴۸ hours (P < ۰.۰۵).CONCLUSION: Resveratrol as an antioxidant can decrease MMP-۹ production, not only by suppressing MMP-۹ expression, but also by augmenting TIMP-۱ production. Altogether, resveratrol as an antioxidant can regulate the MMP-۹/TIMP-۱ balance, and may be considered as a preservative agent in the treatment and prevention of atherosclerosis.
