Rs۲۱۴۱۰۱ Variation of NUCB۲/Nesfatin-۱ Gene: Effects on Metabolic Parameters Independent of Type ۲ Diabetes or Obesity in Coronary Artery Disease

Nesfatin-۱ (NUCB۲) gene, was introduced as a novel satiety factor involves in the control of energy homeostasis in the hypothalamus.Since NUCB۲/nesfatin-۱ has an effect on diabetes and obesity, it is a candidate gene that can be responsible for causing coronaryartery disease (CAD).Therefore, we aimed to identify the association between rs۲۱۴۱۰۱ (C/T) SNP of NUCB۲ gene and the possibleamplified effects of this genetic variation together on the development risk of CAD in this study.This study was carried out in ۱۱۰patients with CAD, and ۶۹ CAD-free controls. The NUCB۲/nesfatin-۱ rs۲۱۴۱۰۱ genotypes were analysed by PCR-RFLP technique. The CCgenotype frequency of the NUCB۲ rs۲۱۴۱۰۱ was greater in control group than CAD group (۳۴.۸% vs.۵۲.۷%; P = ۰,۰۱۹).The CC-genotypewas associated with low serum HDL-C level compared to T allele in female CAD subgroup (P = ۰.۰۳۱), but, not in males. Unlike theCAD group, it was observed that the male control subjects carrying CC genotype have higher serum HDL-cholesterol and diastolicblood pressure levels than those with T allele (P = ۰.۰۴۳ and P = ۰.۰۳۱, respectively) but, not in females. Logistic regression analysisconfirmed that the NUCB۲/Nesfatin-۱ rs۲۱۴۱۰۱ CC-genotype is associated with low serum HDL-cholesterol in femaleCADpatients. Ourresults suggest that the CC genotype of NUCB۲ rs۲۱۴۱۰۱ may contribute to susceptibility CAD risk in relation to low HDL-C and thatthe genetic effect could differ by gender.
