Influence of Extracellular Vesicles from Lactobacillus rhamnosus GG on the Cell Adhesion and mmp۲ and mmp۹ Genes Expression in Colorectal Cancer Cells

Introduction: Lactobacillus rhamnosus GG is a probiotic bacterium with anti-cancer and anti-microbial characteristics. In addition, Extracellular Vesicles (EVs) from Lactobacillus rhamnosus GG showed apoptotic impacts on colorectal cancer cells. In this study, we aimed to isolate EVs from Lactobacillus rhamnosus GG (EVL) and identify its effect on cell adhesion and expression of mmp۲ and mmp۹ genes.Materials and Methods: HT۲۹ cells were exposed to ۱۶, ۳۲, ۶۴, ۱۲۸, ۲۵۶, ۵۱۲, ۱۰۲۴, and ۲۰۴۸ µg/ml EVL, and the cell viability, cell adhesion, and mRNA gene expression were investigated using the MTT assay, cell adhesion assay, and real-time PCR.Results: We found that ۱۲۸, ۲۵۶, ۵۱۲, ۱۰۲۴, and ۲۰۴۸ µg/ml of EVL led to attenuated cell viability of HT۲۹ cells (p<۰.۰۵). Moreover, the expression of both mmp۲ and mmp۹ genes and cell adhesion significantly reduced at ۱۰۲۴ and ۲۰۴۸ µg/ml EVL compared to untreated cells (p<۰.۰۵).Conclusions: According to the findings of the present study, it is suggested that EVL could be effective in cancer cell cytotoxicity.
