Identification of novel genes in different regions of brains: Meta-analysis of schizophrenia transcriptome data

Schizophrenia (SCZ) is a severe mental disorder caused by complex genetic and environmental interplay.Despite a large amount of research was carried out in SCZ, the underlying mechanism is remained to be fullyunderstood. The available drug options for SCZ treatment showed lower than expected efficiency. With theadvent of high-throughput sequencing techniques along with availability of sequencing data, an urgent needto data analysis has appeared. Of note, meta-analysis enables us to integrate several databases and thereforeincrease the reliability of the results. In the present study, we obtained data from different regions of brain,integrate them and introduce new genes associated with SCZ. Microarray gene expression data from ۱۱۹samples (۵۶ normal and ۶۳ SCZ) were retrieved from superior temporal gyrus (STG), hippocampus (HIP),Pre-frontal cortex (PFC) and associative striatum (ASR) following by analysis and integration by GEO۲Rand MetaDE package respectively. Initially, we identify ۴۰۲۶ genes with p-value < ۰.۰۵ using naive sum ofthe ranks method. We then narrowed our list down to ۳۲ genes owing to removing genes having expressionunder |۰.۰۵| in either one of the regions. Furthermore, among the ۳۲ mentioned genes, ۲۷ genes wereinvestigated experimentally in previous studies. Novel genes BOD۱L۱, CHM, EDN۳, ENDOD۱, andANGPTL۴ were found to be differentially expressed genes across all the regions. These candidate genes areof importance for further investigation as they may elucidating the SCZ etiology or being used as biomarkersin the clinical context.
