Neonatal hypothyroidism can cause lifelong mental and physical disorders in humans. Failure to diagnose early can lead to irreparable harm to infants. It can be easily managed with timely detection. Screening programs to diagnose hypothyroidism rely on safety methods in most countries. This method is time consuming, requires laboratory equipment and must be performed by a skilled technician. Therefore, ELISA is not the preferred method for diagnosing neonatal hypothyroidism and can be used as a confirmatory method in infants suspected of having neonatal hypothyroidism. To design a kit, we must first obtain gold-conjugated antibodies. For the synthesis of nanoparticles, a concentration of ۱% in ۱ ml of water was added to ۰.۸ mg of gold nanoparticle powder (HAuCL۴) and heated for ۳ minutes to turn pink. A size analyzer was used to determine the effective diameter of the synthesized gold nanoparticles. Experimentation and pairing with antibodies and evaluation of color and color intensity were evaluated in this study and particle size of ۵۰ nm was considered. Then, using SR۹۵-۱, SR۹۵-۲, and SR۹۵-۳ polyclonal antibodies, which were designed in the previous study against human TSH, along with monoclonal antibodies were used in the kit design. The results of this study showed that these conjugated antibodies can be used to diagnose neonatal hypothyroidism.
